The α-syn amyloid fibril structure shows two protofilaments intertwining along an approximate 2 1 screw axis into a left-handed helix. Each protofilament features a Greek key-like topology. Remarkably, five out of the six early-onset PD familial mutations are located at the dimer interface of the fibril (H50Q, G51D, and A53T/E) or involved in the stabilization of the protofilament (E46K).

They can be formed by essentially any polypeptide, but are of particular interest because of their occurrence in several incurable and debilitating human diseases, e.g. Alzheimer’s disease (AD). A central pathological event in AD is build-up of amyloid fibrils by the amyloid-β (Aβ) peptide. This thesis describes my Amyloid fibrils are homo-molecular protein polymers that play an important role in disease and biological function. While much is known about their kinetics and mechanisms of formation, the origin and magnitude of their thermodynamic stability has received significantly less attention.

It has been demonstrated for Aβ(1-42) peptide and its fragments that their complete structure is inaccessible for the action of proteases, which is an evidence of different ways of association of ring-like oligomers with the formation of fibrils. Many proteins can form amyloid-like fibrils in vitro, but only similar to 30 proteins have been found to cause mammalian amyloid disease, suggesting that

However, structural details of misfolded Aβ(1-42) have remained elusive. Here we present the atomic model of an Aβ(1-42) amyloid fibril, from solid-state NMR (ssNMR) data. It displays triple parallel-β-sheet segments that differ from reported structures of Aβ(1-40) fibrils.

Fibrillar aggregates of the amyloid-β protein (Aβ) are the main component of the senile plaques found in brains of Alzheimer’s disease Identification of regions in the protein chains that form the backbone of amyloid fibril is a direction in the investigation of amyloid formation. Amyloid fibrils can also possess very high kinetic and thermodynamic stabilities, often exceeding those of the functional folded states of proteins (6), as well as a greater resistance to degradation by chemical or biological means (7).

The amyloid fibrils are composed of the amyloid-β peptide (Aβ), a 39–43 amino acid residue peptide produced by cleavage from a larger amyloid precursor protein, APP. The Aβ peptide is known to be present in unaffected individuals and is thought to have a normal physiological role.

The mechanism of fluorescence enhancement upon binding to amyloid has been attributed to the rotational immobilization of the central C More generally, amyloid fibrils are associated with many different human diseases (Knowles et al., 2014), but how mutations alter the propensity of proteins to aggregate into amyloid fibrils is not well understood and there has been no large-scale analysis of the effects of mutations on the formation of any amyloid fibril. Amyloid β (Aβ) monomers are the smallest assembly units, and play an important role in most of the individual processes involved in amyloid fibril formation.

In addition, the Alzheimer's plaque protein Aβ has been shown to undergo reversible amyloid fibril formation to a position of dynamic equilibrium such that reaction thermodynamics can be quantified. Studies of these well-behaved amyloid systems are allowing us to peer more deeply into the process and products of off-pathway misfolding and aggregation.
